The expression levels were compared between control (C) and P. putida BTP1-treated (T) plants. Samples were collected before (0), two days (+2), and four days (+4) after pathogen inoculation. + represents transcripts of the positive control: for TomLoxA, TomLoxB, and TomLoxC, the positive control was RNA extracted from breaker-stage fruit, for TomLoxD and TomLoxF, the positive control was RNA extracted from methyljasmonate-treated plants. Total RNA was extracted from leaves and 20-ÎĽg samples were subjected to RNA-blot analysis, except for the positive controls for TomLoxA, TomLoxB, and TomLoxC, for which 2 ÎĽg was loaded. Transcripts were hybridized with denatured cDNA-specific probes. Quantification of loading of each sample RNA was done by measuring the U.V. fluorescence of ethidium-bromide-stained 28 S rRNA. Loading was found to vary by 20% at most between samples.Back to article page
