The (+)-germacrene A synthase MrTPS3 is assumed to be the key enzyme in matricine biosynthesis converting the ubiquitous precursor farnesyl pyrophosphate into the basic sesquiterpene skeleton. The reactions forming matricine from (+)-costunolide likely include a cyclization, a hydrogenation of a methyl group, two hydroxylations and a subsequent acetylation.
