(A) and (B) Confocal images showing the subcellular localization of ProE7:PIN:GFP (WT-PIN), ProE7:3 m1-PIN:GFP (3 m1-PIN1), and ProE7:M3-PIN:GFP (M3-PIN) in root hair cells after BFA treatment. Transgenic seedlings were pretreated with cycloheximide (50 μM, 30 min), followed by BFA (25 μM, 1 h), and stained with FM4-64 (2 μM). Phosphorylation-defective PIN proteins either co-localized with the BFA compartments (arrow heads) or did not (arrows). Representative images are shown. Bar = 10 μm in all cases.
