GFP-hTalin labels actin microfilaments in hypocotyl epidermis of ArabidopsisConfocal fluorescence images were collected in light-grown seedlings. A) MFs form a network in the cortex of epidermal cells. Image is a maximum projection of 21 optical sections taken at 122 nm intervals along the z-axis. B) MFs form a basket-like structure around a chloroplast in the cell interior. Central image is a single optical section from a stack of 14 images taken at 366 nm intervals. The stack was used to create a three dimensional reconstruction that could be cut into cross sections. Lines labeled XZ and YZ indicate the location of cross sections shown below and to the right of the main image, respectively. GFP-hTalin signal is false-colored green. Chlorophyll fluorescence is false-colored red. In A, DIC image is false-colored blue. Bars: 5 μm.
