Auxin quantification in hypocotyl sections from 19-day old tomatoes. a Cartoon indicating how tissue was collected for auxin determinations. b Tomato hypocotyls were excised at the root-shoot transition zone and transferred onto an agar block before being placed into an enclosed vertical mesh transfer box for 5 h (T5). Control samples were not transferred onto agar blocks, but were instead immediately collected and frozen in liquid nitrogen. Auxin levels in the bottom 0–0.5 cm and upper 2–2.5 cm at the base of the excision were quantified via LC-MS/MS.
